Mechanism of Action
Navitoclax is an orally bioavailable small molecule inhibitor of the B-cell lymphoma-2 (BCL-2) family of proteins. It functions as a dual antagonist of both BCL-2 and BCL-XL, which are key pro-survival proteins often overexpressed in hematologic malignancies. By binding to these proteins, navitoclax prevents them from sequestering pro-apoptotic proteins like BIM and BAK. This restoration of apoptotic signaling triggers mitochondrial outer membrane permeabilization and subsequent programmed cell death in malignant cells.
