Mechanism of Action
Allogeneic, off-the-shelf CD19-directed CAR-NK (chimeric antigen receptor natural killer) cell therapy. NKX019 is manufactured from healthy donor cord blood-derived NK cells engineered to express an anti-CD19 CAR construct along with IL-15 receptor fusion protein (mbIL15) for in vivo persistence. NK cells differ fundamentally from CAR-T cells: they do not recognize host allogeneic antigens via TCR (no HLA mismatch response), thereby eliminating graft-versus-host disease (GvHD) risk. CD19 CAR directs NK cell cytotoxicity specifically toward CD19-expressing malignant B cells.
