Mechanism of Action
Novobiocin inhibits DNA polymerase theta (POLθ/POLQ), an error-prone polymerase essential for microhomology-mediated end joining. In homologous recombination-deficient tumors (e.g., BRCA-mutated), POLθ inhibition is synthetically lethal and can restore sensitivity in PARP-inhibitor-resistant disease.
