Mechanism of Action
Neladalkib (NVL-655) is an oral, brain-penetrant, ALK-selective tyrosine kinase inhibitor engineered to retain potency against a broad spectrum of ALK single and compound resistance mutations (including G1202R and the recalcitrant lorlatinib-resistant compound mutations such as G1202R/L1196M) while sparing the structurally related TRK family. Investigational Indications
- ALK-rearrangement-positive non-small cell lung cancer (NSCLC), including after prior ALK TKIs
- ALK-mutant NSCLC with single or compound resistance mutations
- Other advanced solid tumors with ALK rearrangement or activating ALK mutation
