Mechanism of Action
Oral, selective inhibitor of poly(ADP-ribose) polymerase enzymes PARP1 and PARP2. It blocks PARP-mediated DNA single-strand break repair and exerts potent PARP-trapping, generating cytotoxic double-strand breaks that are lethal to tumors with homologous recombination deficiency (e.g., BRCA1/2 mutations) via synthetic lethality. It is highly brain-penetrant and a strong PARP-trapper relative to some earlier agents.
