Paxalisib — Investigational Agent
PI3K Inhibitor · Phase 2/3 · Kazia Therapeutics
Mechanism of Action
Brain-penetrant, orally available dual inhibitor of class I phosphatidylinositol-3-kinase (PI3K) and mTOR (mTORC1/mTORC2). By blocking the frequently hyperactivated PI3K/AKT/mTOR axis in glioblastoma and crossing the blood-brain barrier, it suppresses tumor-cell proliferation and survival signaling within the CNS.
Investigational Indications
- Newly diagnosed glioblastoma (MGMT-unmethylated)
- Recurrent glioblastoma
- Diffuse midline glioma
- Brain metastases
