Mechanism of Action

Pelareorep is an intravenously administered, non-genetically-modified oncolytic reovirus (Reovirus Type 3 Dearing). It preferentially infects and replicates within tumor cells that have activated RAS signaling, which impairs the antiviral interferon response that normally protects healthy cells — giving the virus selective tropism for transformed cells. Viral replication triggers direct oncolysis and immunogenic cell death, releasing tumor antigens and danger signals that recruit and activate cytotoxic T cells and increase tumor-infiltrating lymphocytes.
