Pumitamig — Investigational Agent
Bispecific Antibody · Phase 3 · BioNTech / Bristol Myers Squibb (originated by Biotheus)
Mechanism of Action
Tetravalent bispecific antibody that simultaneously targets PD-L1 (immune checkpoint) and VEGF-A (pro-angiogenic factor) in a single molecule. By blocking PD-L1 it releases the brake on tumor-directed T cells, while its VEGF-A trap arm normalizes tumor vasculature, reduces immunosuppression in the tumor microenvironment, and enhances immune-cell infiltration. Co-targeting concentrates checkpoint blockade in VEGF-rich tumor tissue, aiming to combine the activity of a PD-(L)1 inhibitor and an anti-angiogenic agent with a single drug and a potentially improved therapeutic index.
