Rinatabart sesutecan — Investigational Agent
Antibody-Drug Conjugate · Phase 2 · Genmab
Mechanism of Action
Rinatabart sesutecan is a folate receptor alpha (FRα)-directed antibody-drug conjugate with a hydrophilic cleavable linker and an exatecan-derived topoisomerase I inhibitor payload. Unlike mirvetuximab soravtansine (a maytansinoid), its topoisomerase I payload and higher drug-to-antibody ratio are designed to retain activity across the full range of FRα expression, including FRα-low tumors. After FRα binding and internalization, the payload induces DNA damage and apoptosis with a bystander effect. This broadens the potentially eligible population in ovarian and endometrial cancers beyond…
Investigational Indications
