S64315 — Investigational Agent
BCL-2 Inhibitor · Phase 1/2 · Servier (with Novartis)
Mechanism of Action
Selective small-molecule inhibitor of myeloid cell leukemia-1 (MCL1), a pro-survival BCL-2 family protein. By binding the BH3-binding groove of MCL1, S64315 releases sequestered pro-apoptotic effectors (BAK/BAX, BIM), restoring intrinsic (mitochondrial) apoptosis in MCL1-dependent malignant cells—a resistance mechanism to BCL-2 inhibition.
