Mechanism of Action

Sigvotatug vedotin is an ADC targeting integrin beta-6 (ITGB6), a cell-surface integrin subunit overexpressed on many epithelial carcinomas including NSCLC. The antibody is conjugated to monomethyl auristatin E (MMAE), a microtubule-disrupting agent, via a protease-cleavable linker. Upon binding integrin beta-6 the ADC is internalized and MMAE is released, causing microtubule disruption, G2/M cell-cycle arrest and apoptosis. Released MMAE also mediates a bystander effect on neighboring tumor cells.
