Mechanism of Action
ABBV-400 (telisotuzumab adizutecan) is a c-Met (MET)-directed antibody-drug conjugate that pairs the anti-c-Met antibody telisotuzumab with a novel proprietary topoisomerase 1 inhibitor payload (Top1i) via a cleavable linker. c-Met is overexpressed in a substantial fraction of colorectal, gastric, and other GI adenocarcinomas and drives proliferation and survival. After binding c-Met, the ADC internalizes and releases its Top1i payload, producing DNA single-strand breaks and apoptosis, with a bystander effect on neighboring cells.
