Tiragolumab — Investigational Agent

Mechanism of Action
Tiragolumab is a fully human IgG1/kappa monoclonal antibody that binds to TIGIT (T-cell immunoreceptor with Ig and ITIM domains). TIGIT is a co-inhibitory receptor that competes with the co-stimulatory receptor CD226 for binding to the ligand PVR (CD155). By blocking TIGIT, tiragolumab prevents the inhibition of T-cells and Natural Killer (NK) cells and may promote the activity of CD226. The IgG1 scaffold also potentially allows for the depletion of TIGIT-expressing regulatory T-cells via antibody-dependent cellular cytotoxicity (ADCC).
