Mechanism of Action
Bispecific antibody that simultaneously blocks DLL4 (Delta-like ligand 4) in the Notch pathway and VEGF-A. VEGF-A drives sprouting angiogenesis while DLL4-Notch signaling regulates functional maturation of tumor vasculature; blocking VEGF alone can upregulate DLL4 as an escape mechanism. Dual blockade produces non-productive, poorly functional tumor vessels and disrupts angiogenic escape, with the goal of deeper and more durable anti-tumor activity than anti-VEGF therapy alone.
Investigational Indications
- Advanced/metastatic biliary tract cancer (cholangiocarcinoma and gallbladder cancer), later-line, with paclitaxel
- Metastatic colorectal cancer (earlier development)
- Other advanced solid tumors
