Mechanism of Action
Trastuzumab duocarmazine is a HER2-targeted antibody-drug conjugate pairing trastuzumab with a cleavable linker-duocarmycin payload (seco-DUBA). After HER2 binding and internalization, proteases release the DNA-alkylating duocarmycin, which binds the minor groove of DNA to cause cell death. The linker also permits payload activation in the tumor microenvironment, enabling activity in HER2-low as well as HER2-positive tumors.
