Mechanism of Action

CD123 x CD3 bispecific antibody (T-cell engager) built on the Xencor XmAb bispecific Fc platform. One arm binds CD123 (IL-3 receptor alpha), which is highly expressed on acute myeloid leukemia blasts and leukemic stem cells but only minimally on normal hematopoietic stem cells; the other arm binds CD3 on T cells. By simultaneously engaging a CD123+ leukemic cell and a cytotoxic T cell, vibecotamab forms an immunological synapse that triggers polyclonal T-cell activation and MHC-independent, targeted killing of CD123-expressing malignant cells.
