Mechanism of Action
Volrustomig is a monovalent bispecific antibody that co-targets PD-1 and CTLA-4 on T cells. It preferentially binds and saturates PD-1 and then, through avidity, directs CTLA-4 blockade selectively to PD-1-high, tumor-infiltrating activated T cells while sparing peripheral CTLA-4-expressing cells. This design aims to deliver the additive antitumor benefit of dual checkpoint inhibition with a potentially improved therapeutic index compared with combining separate anti-PD-1 and anti-CTLA-4 antibodies.
